Description
Palmitoyl-CoA is a coenzyme A derivative of palmitate formed by acyl-CoA synthase. In contrast to medium- and short-chain acyl-CoA, palmitoyl-CoA cannot freely diffuse into the mitochondrial matrix. Formation of palmitoylcarnitine by CPTI is necessary prior to transfer into mitochondria for further fatty acid oxidation (β-oxidation). To study Fatty acid oxidation using Palmitoyl-CoA, Carnitine and low amount of malate is needed on mitochondrial preparations.

Application in HRR
Pa-CoA: Palmitoyl-CoA (Palmitoyl-Coenzyme A lithium salt; Sigma Aldrich, P9716, 5 mg, store at -20°C; FW=1004.94) Preparation of 10 mM stock solution (dissolved in H2O):
- Weigh 5 mg Palmitoyl-CoA.
- Add 497.5 µL H2O.
- Divide into 100 µL portions.
- Store at -20°C.
O2k manual titrations
- Titration volume: 8 µL 10 mM using a 25 µL Hamilton syringe (2 mL O2k-Chamber).
- Final concentration: 40 µM.
- Comments: Please consider that the final concentration needed in the O2k-Chamber may vary according to your cell type and overtitration of Palmitoyl-CoA might inhibit the respiration.
