  • While in Home Office: Gnaigere (talk) 14:03, 19 March 2020 (CET) https://wiki.oroboros.at/index.php/Talk:O2k-Open_Support_alert - Many labs will move to home office mode. In line with our special DL7.4 home office offer (see Agenda today), therefore, I suggest to post in our Agenda analysis-linked O2k-Open Support cases during this time, and collect real-time operation cases for future releases.

Suggestion 1:
O2k-Open SupportWarning: loss of data Can NADH be directly used as substrate in respirometry?

No, you cannot use NADH as a NADH-linked substrate for mitochondrial respiration since NADH is not permeable through the mitochondrial inner membrane (mtIM). The mtIM should be intact for respirometry experiments.
- Nicotinamide adenine dinucleotide

Suggestion 2:

O2k-Open SupportWarning: loss of data Do you perform the leaky chamber test after chamber assembly?

An unnoticed leak from the chamber may affect your data, therefore we recommend performing the Leaky chamber test - especially after chamber reassembly.

Do you know how to select the proper Fluorescence-Sensor and filter set?

Using different Fluorescence-Sensors and filters, the O2k-Fluo Smart-Module allows for measurement of hydrogen peroxide, ATP, mitochondrial membrane potential, calcium, and numerous other applications open for O2k-user innovation.

Do you know how to select the proper fluorescence sensors and filter set?

Do you know how to select the proper fluorescence sensors and filter set?

Using different fluorescence sensors, LED and photodiode filters, the O2k-Fluo Smart-Module (or O2k-Fluo LED2-Module) allows for measurement of hydrogen peroxide, ATP, mitochondrial membrane potential, calcium, and numerous other applications open for O2k-user innovation.
- »Selecting the Smart Fluo-Sensor« and » Selecting a Filter Set«

Arguments:
• It refers generally to both O2k-Fluo Smart-Modul and O2k-Fluo LED2-Modul, therefore, I would write fluorescence sensors.
• The link for Selecting a Filter Set what you put is not appropriate because it goes to a discussion page what we have not published so far, therefore, I would stick to the links in the original alerts.
• The MitoPedia:Fluorometry discussion page does not contain new information. It it summary of information of all fluo-related pages. Therefore, you can find the same information on an other page.
• In the original link the link of Selecting a Filter Set goes to the table with the sensors and filters and not to the listed points.
  • Comments by Caro:
  • Yes, I agree that the content of one of the suggestions which I provided is more appropiated.
  • The question of the entry refers to how to select the appropiate fluorescence sensors and filters. At the end, the first link goes to the section Select the Smart Fluo-Sensors (this is good). The second link does not go to a page which helps with the second question (Selecting a Filter Set), it goes to the O2k-Fluo LED2-Module setup (what about the users with the O2k-FluoRespirometer) and then you can click in another link which goes to a page where some information about the filters can be found inside of the mounting filter section (which is not selecting a filter), and it is not as complete as it is shown in the link Talk:MitoPedia:_Fluorometry#Selecting_a_Filter_Set. Therefore, if I am an user, I do not see the question of the entry fixed. Why the content of the selection of filters is not moved to a front page?
